Overview

ABBV-2001 (also known as ISB 2001) is a first-in-class trispecific T-cell engager (TCE) being developed for the treatment of relapsed or refractory multiple myeloma. It is designed to simultaneously target two antigens on the surface of myeloma cells—B-cell maturation antigen (BCMA) and CD38—while engaging the CD3 epsilon chain on T cells. This dual-targeting approach on the tumor cell is intended to enhance binding avidity and specificity, potentially overcoming resistance mechanisms such as target escape (e.g., loss of BCMA or CD38 expression) and improving efficacy in patients with low antigen density. Developed using the proprietary BEAT (Bispecific Engagement by Antibodies based on the TCR) platform by Ichnos Glenmark Innovation (IGI), ABBV-2001 is currently being evaluated in Phase 1 clinical trials (e.g., TRIgnite-1) both as a monotherapy and in combination with other anti-myeloma agents. AbbVie acquired exclusive rights to the drug in North America, Europe, Japan, and Greater China in 2024.
