ABBV-253 is an investigational antibody-drug conjugate (ADC) developed by AbbVie for the treatment of advanced solid tumors. It consists of a monoclonal antibody targeting the hepatocyte growth factor receptor (c-Met/MET) conjugated to a potent topoisomerase 1 (TOP1) inhibitor payload. c-Met is a cell surface receptor tyrosine kinase that is frequently overexpressed or amplified in various cancers, including non-small cell lung cancer (NSCLC), colorectal cancer, and gastroesophageal cancers, promoting tumor cell proliferation, survival, and metastasis. By binding to c-Met, ABBV-253 is internalized into the tumor cells, where the topoisomerase 1 inhibitor payload is released to induce DNA damage and cell death. ABBV-253 is currently being evaluated in a Phase 1 clinical trial (Study M26-326) for patients with advanced solid tumors expressing c-Met.
