ABBV-257 is a dual-variable domain immunoglobulin G1 monoclonal antibody designed to selectively neutralize both tumor necrosis factor-alpha (TNF-α) and interleukin-17A (IL-17A). It was developed as a bispecific antibody for the treatment of immune-mediated diseases such as rheumatoid arthritis. The drug acts by inhibiting the activity of both TNF-α and IL-17A, two key cytokines involved in inflammatory pathways. Clinical studies demonstrated that ABBV-257 had dose-proportional pharmacokinetics but also showed a high incidence of antidrug antibodies leading to increased drug clearance and reduced exposure. Due to these immunogenicity concerns, development of ABBV-257 has been discontinued[1][2][4].
