Overview

ABBV-428 is a first-in-class, bispecific monoclonal antibody developed by AbbVie for cancer immunotherapy. It targets both mesothelin, a tumor-associated antigen expressed on certain solid tumors, and CD40, an immune co-stimulatory receptor found on antigen-presenting cells. By simultaneously binding to mesothelin and CD40, ABBV-428 provides localized activation of CD40 within the tumor microenvironment. This conditional activation aims to stimulate T-cell infiltration, enhance B-cell activity, and repolarize macrophages toward an anti-tumor phenotype, while minimizing systemic toxicity. The drug acts as a CD40 agonist in the presence of mesothelin-expressing tumor cells, promoting antitumor immune responses. It has been investigated primarily for advanced solid tumors such as ovarian cancer and non-small cell lung cancer in early-phase clinical trials.
