Overview

ABBV-467 is a highly potent and selective small molecule inhibitor of myeloid cell leukemia 1 (MCL-1), a prosurvival B-cell lymphoma 2 family protein critical for tumor maintenance and survival. It was developed by AbbVie as an antineoplastic agent for the treatment of hematologic malignancies, particularly relapsed/refractory multiple myeloma. In preclinical models, ABBV-467 induces rapid apoptosis in MCL-1-dependent tumor cell lines and demonstrates efficacy in xenograft models of human multiple myeloma and acute myelogenous leukemia when used alone or in combination with venetoclax. Clinical trials have shown that while it can achieve disease control, it may also cause increases in cardiac troponin levels—a potential on-target cardiotoxicity associated with MCL-1 inhibition[4][5][8].
