Overview

ABBV-744 is an orally bioavailable, highly selective small molecule inhibitor of the second bromodomain (BDII) of the Bromodomain and Extra-Terminal (BET) family proteins, specifically targeting BRD2, BRD3, and BRD4. It exhibits over 250-fold selectivity for BDII over BDI. The drug was developed by AbbVie as a potential antineoplastic agent for cancers such as acute myeloid leukemia (AML), myelofibrosis, and prostate cancer. Its mechanism involves inhibition of BET proteins that regulate gene expression through recognition of acetylated lysines on histones and other proteins involved in transcriptional activation. Preclinical studies have also shown anti-inflammatory effects via modulation of microglial inflammation pathways[1][2][4][5][6].
