Overview

ABBV-859 is an investigational oral small molecule inhibitor of Tumor Necrosis Factor-Like Ligand 1A (TL1A), currently in Phase 1 development by AbbVie. TL1A (encoded by the TNFSF15 gene) is a cytokine in the TNF superfamily that plays a critical role in the pathogenesis of immune-mediated inflammatory diseases, particularly inflammatory bowel disease (IBD), by promoting both inflammation and fibrosis through its receptor, DR3. By blocking the interaction between TL1A and DR3, ABBV-859 aims to provide a potent anti-inflammatory and anti-fibrotic effect. As an oral small molecule, ABBV-859 represents a potential alternative to injectable monoclonal antibodies targeting the TL1A pathway, offering improved patient convenience and potentially different tissue distribution profiles. It is currently being evaluated for safety, pharmacokinetics, and absolute bioavailability in healthy adult volunteers.
