Overview

ABBV-916 is a human IgG1 monoclonal antibody developed by AbbVie for the treatment of early Alzheimer's disease. It specifically targets and binds to N-terminal truncated, pyroglutamate-modified amyloid beta at amino acid position 3 (AβpE3), a form of amyloid beta implicated in the formation of dense-core plaques in Alzheimer's disease. By binding to AβpE3, ABBV-916 is designed to promote rapid and robust removal of amyloid plaques from the brain, potentially modifying disease progression. Preclinical studies have shown that ABBV-916 reduces plaque burden and recruits microglia without causing microhemorrhage in animal models. The drug was administered intravenously in clinical trials for early-stage Alzheimer's disease but development as a monotherapy has been discontinued due to lack of differentiation from other approved anti-amyloid therapies[2][6][7].
