This combination therapy consists of **ABBV-CLS-484**, a first-in-class, orally bioavailable, small molecule inhibitor of protein tyrosine phosphatase non-receptor type 2 (PTPN2) and protein tyrosine phosphatase non-receptor type 1 (PTPN1), and a **vascular endothelial growth factor receptor tyrosine kinase inhibitor (VEGF TKI)**, a class of antiangiogenic small molecules targeting VEGF signaling. ABBV-CLS-484 is under development as an immunotherapy for advanced solid tumors and is designed to amplify anti-tumor immunity by enhancing interferon signaling and promoting activation of T and NK cells, partly via JAK-STAT pathway augmentation and reduction of T cell exhaustion. The VEGF TKI component, such as cabozantinib, inhibits vascular endothelial growth factor receptor-mediated signaling, which impairs tumor angiogenesis and can further modulate the tumor immune microenvironment. The combination is being evaluated for safety and preliminary efficacy in patients with advanced solid tumors, including renal cell carcinoma, lung cancer, and head and neck cancers[1][2][3][4][5].
