Overview

ABBV-CLS-579 is an orally bioavailable, investigational small molecule inhibitor of protein tyrosine phosphatase non-receptor type 2 (PTPN2) and type 1 (PTPN1), being jointly developed by AbbVie and Calico for the treatment of solid tumors. The drug acts as an antagonist to PTPN2/N1, which are negative regulators of T-cell receptor and cytokine signaling. Inhibition of these enzymes enhances both tumor cell-dependent and immune cell-dependent anticancer mechanisms, including increased interferon signaling via the JAK/STAT pathway, improved tumor antigen presentation, elevated proinflammatory chemokine release, augmented T-cell activation and effector function, and reduced T-cell exhaustion. Clinical trials are ongoing to evaluate its safety and efficacy as monotherapy or in combination with PD-1 inhibitors or VEGF tyrosine kinase inhibitors in patients with advanced solid tumors such as head and neck squamous cell carcinoma (HNSCC), non-small cell lung cancer (NSCLC), and clear cell renal cell carcinoma (ccRCC).
