ABCL635 is a potential first-in-class monoclonal antibody developed as a non-hormonal treatment for moderate-to-severe vasomotor symptoms (VMS), commonly known as hot flashes, associated with menopause. It specifically targets the neurokinin 3 receptor (NK3R), a G-protein-coupled receptor expressed on KNDy neurons in the hypothalamus that regulate body temperature. By antagonizing NK3R, ABCL635 blocks neurokinin B (NKB) signaling, which is implicated in the pathophysiology of VMS during menopause due to estrogen deficiency. This mechanism aims to rebalance neuronal activity and reduce hot flashes without hormonal intervention. The drug is being developed by AbCellera and represents their first clinical-stage program from their GPCR and ion channel platform[1][2][4][5][6].
