aBCMA.BBz CAR-T is an autologous, second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target B-cell maturation antigen (BCMA), a protein highly expressed on the surface of multiple myeloma cells. The CAR construct consists of an anti-BCMA single-chain variable fragment (scFv) linked to a 4-1BB (CD137) costimulatory domain and a CD3ζ signaling domain. Developed and evaluated at the H. Lee Moffitt Cancer Center, this therapeutic candidate has been the subject of research investigating the impact of T-cell source material (e.g., pre-transplant mobilized apheresis vs. post-treatment non-mobilized apheresis) on manufacturing success and product phenotype. Clinical data suggests that CAR-T cells derived from earlier, less-exhausted T-cell populations may exhibit superior expansion and a less differentiated phenotype while maintaining potent cytotoxicity against BCMA-positive myeloma cells.
