ABD-147 is a next-generation targeted radiopharmaceutical biologic therapy developed by Abdera Therapeutics. It is designed to deliver Actinium-225 (225Ac), a highly potent alpha-emitting radioisotope, directly to solid tumors that express delta-like ligand 3 (DLL3) with high affinity. DLL3 is a protein in the Notch signaling pathway and is upregulated on the surface of more than 80% of high-grade neuroendocrine carcinomas—including small cell lung cancer (SCLC) and large cell neuroendocrine carcinoma (LCNEC)—while being absent or rarely expressed on nonmalignant cells. By targeting DLL3-expressing tumor cells with Actinium-225, ABD-147 aims to selectively destroy cancer cells while minimizing radiation exposure to healthy tissue. The drug leverages Abdera’s Radio Optimized Vector Engineering platform for optimized pharmacokinetics and tumor penetration. ABD-147 has received FDA Orphan Drug Designation for neuroendocrine carcinoma and Fast Track designation for extensive-stage SCLC following progression after platinum-based chemotherapy[1][2][4][6][8].
