ABD-1970 is a potent and selective small-molecule inhibitor of monoacylglycerol lipase (MAGL), the primary enzyme responsible for the hydrolysis of the endocannabinoid 2-arachidonoylglycerol (2-AG). By inhibiting MAGL, ABD-1970 increases 2-AG levels, thereby enhancing endocannabinoid signaling at cannabinoid receptors (primarily CB1 and CB2). Preclinical research has demonstrated that ABD-1970 effectively blocks both acute and chronic migraine-associated pain (cephalic allodynia) in mouse models without inducing pharmacological tolerance. The compound was developed by Abide Therapeutics (now part of Lundbeck) and has been used to investigate the role of the endocannabinoid system in modulating migraine mechanisms.
