Drug intelligence / Profile preview
Abemaciclib is an oral **small-molecule cyclin-dependent kinase 4 and 6 inhibitor** developed by Eli Lilly for hormone receptor-positive, HER2-negative breast cancer and other solid-tumor settings under clinical evaluation. By inhibiting CDK4 and CDK6, abemaciclib reduces phosphorylation of retinoblastoma protein, arrests cell-cycle progression from G1 to S phase, and suppresses proliferation of tumor cells that depend on cyclin D-CDK4/6 signaling. It is marketed as Verzenio and is widely used in breast cancer both in advanced/metastatic disease and in adjuvant treatment for high-risk early-stage disease; the provided context also shows ongoing combination-development activity in breast cancer, endometrial cancer, glioblastoma, and other MTAP-deleted solid tumors.
