Overview

A **combination therapy** consisting of abemaciclib (a selective cyclin-dependent kinase 4 and 6 inhibitor), abiraterone acetate (an inhibitor of androgen biosynthesis), and prednisone (a synthetic corticosteroid). This regimen is under investigation for the treatment of metastatic castration-resistant prostate cancer (mCRPC). Abemaciclib inhibits tumor cell proliferation by blocking CDK4/6-mediated cell cycle progression. Abiraterone acetate reduces androgen production by inhibiting CYP17A1, thereby decreasing androgen receptor (AR) activation—a key driver of prostate cancer growth. Prednisone is used to counteract mineralocorticoid excess and provide anti-inflammatory effects. The combination aims to maximize androgen receptor-directed therapy through multi-modal blockade of proliferation and hormonal resistance mechanisms[3][1][2][4][6].
