Overview
**abemaciclib + camizestrant** is a combination of two oral small molecule drugs used in clinical trials for the treatment of estrogen receptor-positive, human epidermal growth factor receptor 2-negative (ER+/HER2−) advanced or early breast cancer. **Abemaciclib** is a selective inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), which blocks cell cycle progression, thereby inhibiting cancer cell proliferation. **Camizestrant** is a next-generation, oral selective estrogen receptor degrader (SERD) and antagonist, promoting estrogen receptor degradation and inhibition of estrogen signaling, a key driver in ER+ breast cancer. The combination has shown to be well-tolerated and potentially efficacious in heavily pretreated patients with ER+/HER2− advanced breast cancer, and is under investigation in Phase 3 trials for early and advanced disease. Both drugs are administered orally. Camizestrant is under development by AstraZeneca[1][3][5].
