This combination therapy regimen, studied at Guizhou Provincial People's Hospital, consists of the CDK4/6 inhibitor abemaciclib (Verzenio) administered alongside endocrine therapy and ovarian function suppression (OFS). Abemaciclib is a small molecule that selectively inhibits cyclin-dependent kinases 4 and 6 (CDK4/6), which are critical for the transition of cells from the G1 to the S phase of the cell cycle. By blocking these kinases, abemaciclib inhibits the proliferation of hormone receptor-positive (HR+) breast cancer cells. The addition of endocrine therapy (such as aromatase inhibitors or tamoxifen) and OFS (typically GnRH agonists) serves to further deplete estrogen levels or block estrogen receptor signaling, providing a synergistic anti-tumor effect. This specific regimen is being evaluated in a Phase 1 clinical trial to determine its safety and tolerability when used concurrently with postoperative radiotherapy in patients with HR+/HER2- high-risk breast cancer.
