Abemaciclib + xentuzumab is a combination therapy consisting of abemaciclib, a CDK4/6 inhibitor, and xentuzumab, an insulin-like growth factor (IGF) ligand-neutralizing antibody. This combination targets two complementary pathways involved in cancer cell proliferation. Abemaciclib disrupts cell cycle progression by inhibiting CDK4/6, while xentuzumab neutralizes IGF ligands, preventing activation of the IGF pathway which leads to increased cyclin D1 expression. The dual inhibition approach aims to enhance anti-tumor efficacy, particularly in hormone receptor-positive (HR+), HER2-negative breast cancer.
