Overview

Abeotaxane (TPI 287) is a third-generation taxane derivative developed by Cortice Biosciences (formerly Tapestry Pharmaceuticals) for the treatment of various cancers and neurodegenerative diseases. Unlike first-generation taxanes such as paclitaxel, abeotaxane is designed to circumvent multidrug resistance (MDR) by not acting as a substrate for the P-glycoprotein (P-gp) efflux pump. This characteristic allows it to achieve higher intracellular concentrations in resistant tumor cells and effectively cross the blood-brain barrier. Its mechanism of action involves binding to and stabilizing microtubules, which induces cell cycle arrest at the G2/M phase and leads to apoptosis. In oncology, it has been investigated for advanced solid tumors, including glioblastoma and pancreatic ductal adenocarcinoma, particularly in patients who have progressed on standard therapies like gemcitabine.
