Overview

Abequolixron + docetaxel is a combination therapy being investigated for the treatment of various cancers, particularly lung cancer. This combination pairs abequolixron, an orally bioavailable liver X receptor (LXR) agonist, with docetaxel, a standard chemotherapy agent. Abequolixron (also known as RGX-104) is a first-in-class small molecule that selectively targets and activates the liver X receptor beta (LXRbeta/NR1H2), which leads to the transcription of tumor suppressor genes, particularly apolipoprotein E (ApoE). This activation enhances anti-tumor immunity through multiple mechanisms: - Inhibiting tumor angiogenesis (blood vessel formation) - Depleting immunosuppressive myeloid-derived suppressor cells (MDSCs) - Activating cytotoxic T lymphocytes (CTLs) - Reversing immune evasion in the tumor microenvironment When combined with docetaxel (a taxane chemotherapy), this regimen has shown promising clinical activity in early trials, particularly in lung cancer patients.
