Overview

Abetimus sodium is a synthetic immunosuppressant and tolerogen developed for the treatment of systemic lupus erythematosus (SLE), specifically lupus nephritis. It is a tetrameric oligonucleotide conjugate composed of four double-stranded oligodeoxyribonucleotides attached to a nonimmunogenic carrier platform. The drug was designed to induce tolerance in B cells by cross-linking surface antibodies directed against double-stranded DNA (dsDNA), which are implicated in the pathogenesis of lupus nephritis. By binding to these anti-dsDNA antibodies both in circulation and on B cell surfaces, abetimus forms immune complexes that reduce antibody titers and may disable pathogenic B cells[1][3][4][5][6]. Despite showing reductions in anti-dsDNA antibody levels and some clinical benefit in reducing renal flares, pivotal phase III trials failed to demonstrate significant prolongation of time to renal flare compared with placebo[1][8].
