ABI-013 (also known as nab-CY196) is a solvent-free, nanoparticle albumin-bound (nab) formulation of CY196, a novel docetaxel analog. Developed using proprietary nab-technology originally by Abraxis BioScience (later acquired by Celgene and subsequently Bristol Myers Squibb), ABI-013 is designed to improve the therapeutic index of docetaxel-based chemotherapy by avoiding systemic toxicities associated with conventional solvent vehicles (such as polysorbate 80 and ethanol). As a microtubule stabilizer, the active taxane moiety binds to tubulin, inhibiting microtubule depolymerization and arresting cell division. Preclinical studies have demonstrated that ABI-013 possesses superior antitumor activity and a favorable safety profile compared to docetaxel in xenograft models of breast, colorectal, and non-small cell lung cancers.
