Overview

ABI-171 is a novel small molecule fluoro-catechin derivative developed by Avanti Biosciences for the treatment of various fibrotic diseases. It acts as a dual inhibitor of dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A) and proviral integration site for Moloney murine leukemia virus 1 (PIM1) kinases. These targets are key regulators in the signaling pathways that drive fibrotic tissue formation and inflammation. ABI-171 has demonstrated significant antifibrotic activity in preclinical models of idiopathic pulmonary fibrosis (IPF) and liver fibrosis, including nonalcoholic steatohepatitis (NASH). The drug has received FDA Orphan Drug Designation for the treatment of IPF and is currently undergoing IND-enabling studies, with a Phase 1 clinical trial planned to commence in 2026.
