ABI-4334 is a highly potent, next-generation small molecule capsid assembly modulator (CAM) developed by Assembly Biosciences for the treatment of chronic hepatitis B virus (HBV) infection. It acts as a core protein inhibitor, interfering with the formation and function of the HBV capsid—the shell that surrounds and protects the viral genetic material. By targeting the HBV core protein, ABI-4334 inhibits viral replication through two main mechanisms distinct from nucleos(t)ide reverse transcriptase inhibitors or interferon-alpha therapies: it blocks HBV DNA replication and prevents formation of new covalently closed circular DNA (cccDNA), which serves as a persistent reservoir for HBV in infected cells. Preclinical studies show that ABI-4334 accelerates empty capsid assembly and can prematurely disrupt intact capsids, including those containing DL-DNA, potentially impacting viral integration into host genomes. Interim Phase 1b clinical trial data demonstrate strong antiviral activity in adults with chronic hepatitis B, with significant reductions in both plasma HBV DNA and RNA levels after 28 days of oral dosing[1][2][3][5][6][8].
