ABI-6250 is an investigational, orally bioavailable small molecule entry inhibitor developed for the treatment of chronic hepatitis delta virus (HDV) infection. It acts by selectively inhibiting the sodium taurocholate cotransporting polypeptide (NTCP), a bile acid transporter that serves as the entry receptor for both HDV and hepatitis B virus (HBV) into liver cells. By blocking NTCP, ABI-6250 prevents viral entry and subsequent infection of hepatocytes. Preclinical studies have demonstrated low nanomolar potency against multiple HDV genotypes, high selectivity for NTCP over other bile acid transporters, and a pharmacokinetic profile supportive of once-daily oral dosing. The drug is currently in Phase 1 clinical trials to evaluate its safety, tolerability, pharmacokinetics, and effects on serum bile acids in healthy volunteers.
