ABI-H3733 is an orally administered small molecule antiviral drug developed by Assembly Biosciences for the treatment of chronic hepatitis B virus (HBV) infection. It acts as a next-generation core inhibitor targeting the viral core protein of HBV. The drug has demonstrated increased potency against both covalently closed circular DNA (cccDNA) formation and new virus production compared to first-generation core inhibitors in preclinical studies. Clinical trials have shown that it can significantly reduce plasma HBV DNA levels in patients with chronic hepatitis B. The primary goal is to provide more durable off-treatment virologic responses than current standard-of-care therapies such as nucleos(t)ide reverse transcriptase inhibitors[1][2][5][6][7].
