Abicipar pegol is a long-lasting, PEGylated designed ankyrin repeat protein (DARPin) that binds with high affinity to vascular endothelial growth factor A (VEGF-A), inhibiting its activity. By blocking VEGF-A, it prevents abnormal blood vessel growth and leakage in the eye, which are key drivers of neovascular age-related macular degeneration (nAMD) and diabetic macular edema (DME). Abicipar was developed as an intravitreal injection for these retinal diseases. Despite demonstrating non-inferior efficacy to ranibizumab in clinical trials with less frequent dosing, its development was hampered by a higher rate of intraocular inflammation compared to existing therapies. The drug was developed by Molecular Partners and Allergan (now part of AbbVie), but has not been approved due to safety concerns[2][3][5][6].
