Overview

Abietic acid is a naturally occurring abietane diterpenoid and the principal resin acid found in rosin from coniferous trees, especially pines. It features a tricyclic structure with two alkene groups and a carboxylic acid moiety. Abietic acid serves as a plant metabolite, likely functioning to protect trees from insect attack and wounds. Commercially, it is used in the manufacture of lacquers, varnishes, soaps, adhesives, cosmetics, soldering fluxes, and as an intermediate in pharmaceuticals. Pharmacologically, abietic acid exhibits anti-inflammatory and immunomodulatory effects—acting as an inhibitor of testosterone 5α-reductase (in vitro), modulating NF-κB and MAPK/AP-1 pathways via PPARα/γ activation (anti-inflammatory), suppressing age-related inflammation and improving skin photoaging. It has demonstrated anticancer activity through mechanisms including inhibition of NF-κB signaling; PI3K/AKT/mTOR pathway modulation; cell cycle arrest at G0/G1 phase; induction of apoptosis via mitochondrial-dependent/extrinsic pathways; AMPK pathway activation; ferroptosis induction; inhibition of MMPs/u-PA secretion (anti-metastatic); regulation of Th17/Treg balance (immunomodulation); reduction of proinflammatory cytokines such as TNF-α and IL-17A; modulation of gut microbiota composition in animal models[2][5][7][9]. Abietic acid is approved by the US FDA for use as a miscellaneous food additive.
