Overview

Abiraterone acetate is an orally active, small molecule inhibitor of the enzyme cytochrome P450 17A1 (CYP17A1), which possesses both 17α-hydroxylase and 17,20-lyase activities. This enzyme is a critical catalyst in the biosynthesis of androgens, and its inhibition leads to the suppression of testosterone production in the testes, adrenal glands, and prostate tumor tissue. Developed by Cougar Biotechnology (a subsidiary of Johnson & Johnson), abiraterone acetate is indicated for the treatment of metastatic castration-resistant prostate cancer (mCRPC) and metastatic high-risk castration-sensitive prostate cancer (mHSPC). In the context of the COU-AA-206 Phase 1b clinical trial (NCT01400555), 'Cohort 2' specifically refers to the abiraterone acetate component being evaluated in combination with docetaxel and prednisone to determine the safety and recommended dose for chemotherapy-naïve patients.
