Abiraterone acetate + enzalutamide refers to the alternative hormonal therapy intervention arm in the Phase 3 clinical trial NCT04139772, sponsored by the National Cancer Institute, Naples. Abiraterone acetate is a small molecule inhibitor of the CYP17A1 enzyme, which is critical for androgen biosynthesis in the testes, adrenal glands, and prostate tumor tissue; it is typically administered with prednisone to manage mineralocorticoid excess. Enzalutamide is a small molecule androgen receptor (AR) antagonist that competitively inhibits androgen binding, AR nuclear translocation, and DNA binding. The trial aims to determine if sequential hormonal therapy is a viable alternative to chemotherapy in the second-line setting for asymptomatic or oligosymptomatic mCRPC patients.
