Overview

Abiraterone acetate + methylprednisolone is a combination therapy used primarily for the treatment of metastatic castration-resistant prostate cancer (mCRPC) and metastatic high-risk castration-sensitive prostate cancer (CSPC). Abiraterone acetate is a prodrug of abiraterone, which acts as a selective and irreversible inhibitor of the enzyme steroid 17-hydroxylase/17,20 lyase (CYP17A1), thereby blocking androgen biosynthesis that fuels prostate cancer growth. Methylprednisolone, a synthetic glucocorticoid corticosteroid, is co-administered to mitigate mineralocorticoid-related side effects such as hypertension, hypokalemia, and fluid retention caused by CYP17A1 inhibition. The combination improves survival in patients with advanced prostate cancer who have progressed despite androgen deprivation therapy[2][3][5][8].
