Drug intelligence / Profile preview
Abiraterone Acetate Tablets (II) is an improved nanocrystal formulation of abiraterone acetate, a small molecule prodrug that inhibits the enzyme CYP17 (17α-hydroxylase/C17-20 lyase). This enzyme is a critical component of the androgen biosynthesis pathway in the testes, adrenal glands, and prostate tumor tissues. This specific formulation, developed by Ding-Wei Ye and colleagues, utilizes nanocrystal technology and the absorption enhancer Sodium N-[8-(2-hydroxy-benzoyl) amino] caprylate (SNAC) to improve oral bioavailability and reduce pharmacokinetic variability. These improvements allow for a significantly lower therapeutic dose (300 mg daily) compared to the 1000 mg daily dose required for the originator product (Zytiga), while maintaining equivalent efficacy in treating metastatic castration-resistant prostate cancer (mCRPC). The formulation also minimizes the food effect, allowing for administration without dietary restrictions.
