Abiraterone decanoate (ASP5541, PRL-02) is a long-acting prodrug of abiraterone, a potent inhibitor of the enzyme CYP17A1 (17α-hydroxylase/C17,20-lyase). Developed by Astellas Pharma, it is designed as a sustained-release formulation for the treatment of prostate cancer. By inhibiting CYP17A1, the drug blocks the biosynthesis of androgens in the testes, adrenal glands, and prostate tumor tissue, thereby depriving androgen-sensitive prostate cancer cells of the hormones required for growth. The decanoate ester formulation aims to provide a longer duration of action compared to standard abiraterone acetate, potentially allowing for less frequent dosing via intramuscular injection. It is currently in Phase 2 clinical development.
