Overview

ABK-EGFR-1 is a novel, fourth-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) developed by Abbisko Therapeutics. It is specifically designed to target the EGFR C797S resistance mutation, which frequently emerges in non-small cell lung cancer (NSCLC) patients following treatment with third-generation TKIs like osimertinib. The drug is characterized by high selectivity for the C797S mutation over wild-type EGFR, potentially minimizing off-target toxicities such as skin rash and diarrhea. Preclinical studies presented at the 2026 AACR Annual Meeting demonstrated significant antitumor activity in multiple C797S-driven xenograft models (including Del19/T790M/C797S and L858R/C797S) and excellent blood-brain barrier penetration, suggesting potential efficacy against brain metastases.
