ABK-GIPR-1

ABK-GIPR-1 is a highly potent, orally available small-molecule antagonist of the glucose-dependent insulinotropic polypeptide receptor (GIPR). Developed by Abbisko Therapeutics, it is being investigated for the treatment of obesity and related metabolic disorders. Preclinical studies in diet-induced obesity (DIO) mouse models have demonstrated that ABK-GIPR-1 significantly enhances the weight-reducing efficacy of GLP-1 receptor agonists (GLP-1RAs), such as semaglutide and orforglipron. The combination of ABK-GIPR-1 and semaglutide resulted in over 40% body weight loss, primarily through fat mass reduction, while also improving hepatic lipid accumulation, blood lipid profiles, and glucose metabolism.
