ABL101

ABL101 (also known as TJ-L101) is an investigational bispecific antibody designed to target B-cell maturation antigen (BCMA) and the co-stimulatory receptor 4-1BB (CD137). Developed through a collaboration between ABL Bio and I-Mab Biopharma, the molecule utilizes ABL Bio's Grabody-T platform to facilitate conditional 4-1BB activation. This mechanism ensures that the T-cell stimulatory signal is only triggered upon the antibody's binding to BCMA-expressing tumor cells, thereby localizing the immune response and minimizing systemic toxicities, such as hepatotoxicity, which are commonly associated with systemic 4-1BB agonism. ABL101 is primarily being developed for the treatment of hematologic malignancies, most notably multiple myeloma, where BCMA is highly and specifically expressed. The drug is currently in the IND-enabling stage of development.
