Overview

ABL102 is a first-in-class bispecific antibody developed by ABL Bio using its proprietary Onabody platform. It is designed to simultaneously target Receptor Tyrosine Kinase Like Orphan Receptor 1 (ROR1) and the costimulatory receptor 4-1BB (CD137). The drug's mechanism of action involves ROR1-dependent 4-1BB clustering, which ensures that T-cell activation occurs specifically within the tumor microenvironment where ROR1 is overexpressed, thereby reducing the risk of systemic toxicity typically associated with 4-1BB agonists. Furthermore, ABL102 facilitates the depletion of intratumoral regulatory T cells (Tregs) through an intact Fc region via FcγRI-mediated mechanisms. Preclinical studies have demonstrated potent antitumor activity and a favorable safety profile. It is currently in Phase 1 clinical development for the treatment of ROR1-positive solid tumors and hematologic malignancies.
