Overview

ABL104 (also known as YH32364) is a bispecific antibody designed to target both the Epidermal Growth Factor Receptor (EGFR) and 4-1BB (CD137). It is engineered using an IgG1 isotype and developed to overcome EGFR drug resistance in cancer by combining tumor-directed 4-1BB agonism with EGFR signal blockade. By binding to EGFR on tumor cells and activating 4-1BB on T cells within the tumor microenvironment, ABL104 blocks proliferative signaling in cancer cells while simultaneously stimulating anti-tumor immune responses. Preclinical studies have shown that it induces T cell activation and IFN-γ secretion leading to tumor cell lysis, with superior efficacy compared to cetuximab in animal models. The drug has demonstrated a favorable safety profile in non-human primate studies. Its development focuses primarily on solid tumors that overexpress EGFR or are resistant to existing EGFR-targeted therapies[1][2][4].
