Overview

ABL105 (also known as YH32367) is a bispecific monoclonal antibody developed for the treatment of solid tumors that overexpress human epidermal growth factor receptor 2 (HER2), including HER2-positive breast cancer, gastric cancer, gastroesophageal junction carcinomas, and biliary tract cancer. The drug is designed to simultaneously target HER2 and Tumor Necrosis Factor Receptor Superfamily Member 9 (4-1BB or CD137). By binding to both targets, ABL105 acts as a T cell engager—activating T cells specifically within the tumor microenvironment through tumor-directed 4-1BB agonism. This mechanism aims to enhance anti-tumor immune responses while minimizing systemic toxicity. Preclinical studies have shown that ABL105 induces IFN-γ secretion and elicits tumor cell death in co-culture conditions. It has demonstrated superior efficacy in activating immune cells and inhibiting tumor growth compared to equimolar dosing of trastuzumab and benchmark 4-1BB agonistic antibodies in animal models. Clinical development is ongoing with Phase I/II trials evaluating its safety and anti-tumor activity in patients with HER2-positive locally advanced or metastatic solid tumors[1][2][3][4][5][7].
