Overview

ABL301 is a bispecific antibody designed to cross the blood-brain barrier (BBB) and selectively target aggregated forms of alpha-synuclein (SNCA), a protein implicated in the pathogenesis of Parkinson’s disease and multiple system atrophy. The drug combines an anti-alpha-synuclein IgG with an anti-insulin-like growth factor 1 receptor (IGF1R) single-chain variable fragment (scFv), which acts as a BBB shuttle. This design enables improved delivery of the therapeutic antibody into the brain via receptor-mediated transcytosis. Mechanistically, ABL301 facilitates microglial phagocytosis of extracellular aggregated alpha-synuclein and inhibits its cell-to-cell transmission—two processes believed to contribute to neurodegeneration in synucleinopathies. Developed by ABL Bio, it is currently in Phase 1 clinical development for Parkinson’s disease[1][2][3][5][6].
