ABL501 is a bispecific antibody designed to simultaneously block the immune checkpoint proteins LAG-3 (Lymphocyte Activation Gene-3) and PD-L1 (Programmed Death Ligand 1). By targeting both pathways, ABL501 releases T cells from tumor-mediated suppression, leading to enhanced T cell activation and antitumor responses. Preclinical studies have shown that ABL501 induces stronger cytotoxic activity against tumor cells compared to single-agent therapies or combinations of separate LAG-3 and PD-L1 antibodies. The drug is being developed primarily for the treatment of advanced or metastatic solid tumors, with ongoing Phase 1 clinical trials evaluating its safety, tolerability, and preliminary efficacy[1][4][7]. Developed using Grabody-I platform technology by ABL Bio, it represents a novel approach in cancer immunotherapy aimed at overcoming resistance seen with conventional PD-(L)1 therapies[6].
