ABM-05X

ABM-05X is an investigational monoclonal antibody developed for the local treatment of tenosynovial giant cell tumor (TGCT), a rare, locally aggressive neoplasm affecting synovial joints. It is designed to be administered via intra-articular injection directly into the affected joint, aiming to achieve high local drug concentrations at the tumor site while minimizing systemic exposure and associated adverse events. The mechanism of action involves targeting and inhibiting colony-stimulating factor 1 receptor (CSF1R), which plays a key role in regulating macrophages and related cells implicated in TGCT pathogenesis. By blocking CSF1R signaling—mediated by its ligands CSF1 and IL-34—the drug disrupts recruitment and proliferation of tumor-associated macrophages, leading to tumor shrinkage. ABM-05X has received FDA fast track designation for TGCT of the knee based on promising phase 2 data showing clinical benefit with minimal toxicity[7][9][10]. It is also being investigated for potential use in colorectal cancer patients with minimal residual disease[6].
