ABM-1310 + cobimetinib is a combination therapy of two small molecules targeting the MAPK pathway for the treatment of advanced or metastatic solid tumors harboring BRAF mutations, particularly BRAF V600[1][2][3][5]. ABM-1310 is a novel, highly potent and selective BRAF inhibitor designed for high brain penetration and cell permeability[1][6][7]. Cobimetinib is a MEK inhibitor that blocks MEK1/2 signaling. Together, they are being investigated for their synergistic efficacy against tumors with BRAF V600 mutations—especially central nervous system (CNS) tumors, melanoma, thyroid cancer, pancreatic cancer, and brain metastases—after failure of prior BRAF ± MEK therapy[1][2][3][5]. Preliminary phase 1 clinical data indicate reasonable tolerability and initial signs of activity in several tumor types, including glioblastoma and pleomorphic xanthoastrocytoma[1][5]. ABM-1310 is developed by ABM Therapeutics and cobimetinib by Genentech/Roche.
