Overview

ABM-168 is a novel small molecule allosteric inhibitor of MEK1 and MEK2 (mitogen-activated protein kinase kinase 1/2), developed by ABM Therapeutics. It is designed to block the MAPK signaling pathway, which is frequently activated in cancers with RAS or RAF mutations. The drug demonstrates high water solubility, cell permeability, and excellent blood-brain barrier penetration—making it particularly promising for treating brain metastases and primary brain tumors. Preclinical studies have shown strong anti-cancer activity both as monotherapy and in combination with other agents across multiple cancer models. Currently, ABM-168 is being evaluated in Phase 1 clinical trials for advanced/metastatic solid tumors—including those with RAS/RAF/NF1 mutations—with a focus on patients who have brain metastases or primary CNS tumors[2][4][6][8].
