Overview

ABM-2752 is a potent and selective small molecule inhibitor of HPK1 (hematopoietic progenitor kinase 1), developed by ABM Therapeutics. HPK1 is a serine/threonine kinase that acts as a negative regulator of T-cell activity; thus, inhibition of HPK1 can enhance immune responses against tumors. Preclinical studies have shown that ABM-2752 demonstrates strong antitumor efficacy in multiple syngeneic mouse models (including CT26, MC38, and B16F10), both as monotherapy and in combination with PD-1 antibody checkpoint inhibitors. The compound has also exhibited favorable drug-like properties in ADME and PK studies and has shown the ability to induce IL-2 release while inhibiting SLP-76 phosphorylation in T-cell activation assays. As of the latest updates, it remains in preclinical/IND-enabling development for cancer indications[1][2][3][5][6].
