Overview

ABM-3249 is a potent and selective small molecule inhibitor of cyclin-dependent kinase 8 (CDK8), developed by ABM Therapeutics. It demonstrates high selectivity for CDK8 with an IC50 of 1.3 nM and limited activity against other CDK kinases (49–100 times less potent). Mechanistically, it inhibits the phosphorylation of downstream molecules such as STAT1, impacting gene transcription pathways involved in tumor cell invasion, metastasis, and drug resistance. In preclinical cancer models—including subcutaneous MV4-11 leukemia and MC38 colon cancer—ABM-3249 has shown significant anti-tumor efficacy both as a monotherapy and in combination with PD-1 antibodies. The compound also exhibits favorable safety profiles in preliminary tolerance studies. Its development is focused on oncology indications such as adult acute myeloblastic leukemia and colonic cancer[1][2][5].
