ABM-4095 is a novel, potent, orally available small molecule inhibitor targeting phosphorylated MEK (pMEK), developed by ABM Therapeutics. It is designed to inhibit the MEK1/2 kinases within the MAPK signaling pathway, which is frequently reactivated in RAS-mutant cancers and contributes to drug resistance against other targeted therapies. Preclinical studies have demonstrated that ABM-4095 has high cell permeability and significant anti-tumor activity both in vitro and in vivo across several RAS-mutant cancer models, including pancreatic cancer, lung cancer (notably KRAS-mutated non-small cell lung cancer), and colorectal cancer. The compound shows dose-dependent tumor inhibition as a single agent and exhibits synergistic effects when combined with other agents such as KRAS G12C inhibitors (e.g., AMG-510) or EGFR inhibitors (e.g., erlotinib). Its improved brain permeability also positions it for potential use against brain metastases[1][2][4][8].
