ABN202 is an antibody-cytokine fusion protein (ACFP) designed to overcome the limitations of traditional type I interferons (IFNs) in cancer therapy. It consists of a glycoengineered IFN-β mutein, which exhibits improved biophysical properties and retained functional activity under acidic tumor microenvironment (TME) conditions, fused to a TROP2-targeting monoclonal antibody. This design enables tumor-selective delivery of the IFN-β mutein, enhancing its anti-tumor efficacy while improving the preclinical toxicity profile compared to other type I IFNs. ABN202 induces potent direct tumor cell killing and enhances immune cell-mediated cytotoxicity by activating type I IFN signaling.
