ABO2101 is a novel mRNA-encoded therapeutic cancer vaccine developed by Abogen Bioscience, specifically targeting Human Papillomavirus type 16 (HPV16) E6 and E7 oncoproteins. The vaccine is engineered with a unique antigen-expressing cassette that incorporates endolysosomal trafficking domains and antigen-presenting cell (APC) maturation stimulators. This design is intended to enhance both Class I and Class II antigen presentation and promote APC maturation, thereby inducing a robust T cell-mediated adaptive immune response. Preclinical data presented at AACR 2024 demonstrated that ABO2101 can eradicate established TC-1 tumors in murine models and shows synergistic anti-tumor activity when combined with immune checkpoint inhibitors such as PD-1 and CTLA-4 antibodies. The vaccine is being developed for the treatment of HPV-associated cancers, including cervical, anogenital, and oropharyngeal malignancies.
